Isolation and Detection of Antibiotic-Resistant Staphylococcus aureus from Street-Vended Chicken Intestines in a Philippine Urban Setting

Street food has become popular worldwide due to its convenience, affordability, and diverse flavors, such as isaw and tomato sauce. However, foodborne pathogens, especially Staphylococcus aureus are commonly associated with street food. This study aimed to detect putative antibiotic-resistant Staphylococcus aureus in isaw and tomato sauce randomly collected from the food stalls around Barangay Ampayon, Butuan City. The putative Staphylococcus aureus isolates were selectively grown and characterized morphologically using Tryptone Soy Broth and Baird-Parker Agar. Putative Staphylococcus aureus isolates were further characterized through its biochemical reaction to assays such as Gram staining, indole test, catalase test, methyl-red test, triple sugar iron test, and coagulase test. The study revealed that 19 isolates of presumptive Staphylococcus aureus were isolated, and only 15 follows with the characteristics of a typical Staphylococcus aureus through biochemical tests. Moreover, an antibiotic susceptibility test was conducted using the disc diffusion method on Mueller Hinton Agar with antibiotics such as oxacillin, gentamicin, ciprofloxacin, tetracycline, and cotrimoxazole. All 15 suspected Staphylococcus aureus isolates were resistant to the oxacillin, while 93.33%, 100%, 93.33%, and 100% were susceptible to gentamicin, ciprofloxacin, tetracycline, and cotrimoxazole, respectively. One isolate was found to be resistant to three different classes of antibiotics: oxacillin, gentamicin, and tetracycline indicating that the isolate is multi-drug resistant (MDR). Thus, this study found antibiotic-resistant Staphylococcus aureus with one isolate being an MDR strain, suggesting health risks to both vendors and consumers of these food samples.
